Characterization of interphase nuclei from triploid hybrids between Pennisetum purpureum Schumach and Pennisetum glaucum (L.) R. Br. Ciência e Agrotecnologia
Fajardo,Cristiane Gouvêa; Davide,Lisete Chamma; Pereira,Antônio Vander.
This study evaluated the structure and the volume of interphase nuclei from root meristems of the genotypes of napier grass (Pennisetum purpureum), pearl millet (Pennisetum glaucum) and hybrids resultant of such breeding. In napier grass, nuclei were areticulate. Both pearl millet and the triploid hybrid had semi-reticulate nuclei; also, the hybrid presented a small proportion (6%) of areticulate nuclei. Pearl millet had the highest averages of nuclear dimensions, such as volume, diameter and radius, followed by the interspecific hybrid and napier grass. There was no intraspecific variation for the type of nuclear structure, which indicates this feature is important for cytotaxonomic studies involving the genus Pennisetum. Results demonstrated that...

Cover crops and herbicide timing management on soybean yield under no-tillage system PAB
Nascente,Adriano Stephan; Crusciol,Carlos Alexandre Costa.
The objective of this work was to evaluate the effect of cover crops and timing of pre-emergence herbicide applications on soybean yield under no-tillage system. The experiment consisted of four cover crops (Panicum maximum, Urochloa ruziziensis, U. brizantha, and pearl millet) and fallow, in addition to four herbicide timings (30, 20, 10, and 0 days before soybean sowing), under no-tillage system (NTS), and of two control treatments under conventional tillage system (CTS). The experimental design was a completely randomized block, in a split-plot arrangement, with three replicates. Soybean under fallow, P. maximum, U. ruziziensis, U. brizantha, and pearl millet in the NTS and soybean under U. brizantha in the CTS did not differ significantly regarding...

Cytogenetics of Pennisetum purpureum Schumack x Pennisetum glaucum L. hybrids and their parents Ciência e Agrotecnologia
Barbosa,Sandro; Davide,Lisete Chamma; Pereira,Antônio Vander.
Cytogenetic studies were carried out on five accesses of interspecific hybrids of Pennisetum purpureum and Pennisetum glaucum and their parentals, which are part of the Forage Germplasm Bank of the "Centro Nacional de Pesquisa de Gado de Leite" of EMBRAPA, Brazil. The mitotic cytogenetic analysis, performed using stain by Schiff reactive, confirmed the chromosome number of 2n = 14, 28 and 21 for the accesses of P. glaucum, P. purpureum and their hybrids, respectively. For each access, descriptions of the chromosome complement and idiograms are presented which allowed to establish the karyotypic relationships between the hybrids and their parentals. Based on the total chromosomal length data it was possible to establish the karyotype classification of the...

Genetic analysis of reciprocal differences in the inheritance of in vitro characters in pearl millet Genet. Mol. Biol.
Satyavathi,Valluri V; Manga,V.; Rao,Muktinutalapati V. Subba; Chittibabu,Malladi.
Abstract Reciprocal differences persist in nature because of the unequal contribution of cytoplasmic determinants from male and female gametes to the zygote. The inheritance of genetic differences is an important factor that influences various traits, including somatic embryogenesis and regeneration in vitro. In this report, we estimate the cytoplasmic and maternal effects in pearl millet and their adequacy in describing the observed reciprocal differences based on an in depth study of the parents, F2s and reciprocal backcross progenies needed for fitting genetical models. Our study revealed that of the two characters examined, embryogenic callus quantity and regeneration frequency, the former showed a greater proportion of cytoplasmic nuclear interaction...

Genetic architecture of purple pigmentation and tagging of some loci to SSR markers in pearl millet, Pennisetum glaucum (L.) R. Br. Genet. Mol. Biol.
Varalakshmi,Pusapati; Tavva,Surya S Mohan Dev; Rao,Palakollu V Arjuna; Rao,Muktinutalapati V Subba; Hash,Charles T.
This report describes the construction of integrated genetic maps in pearl millet involving certain purple phenotype and simple sequence repeat (SSR) markers. These maps provide a direct means of implementing DNA marker-assisted selection and of facilitating "map-based cloning" for engineering novel traits. The purple pigmentation of leaf sheath, midrib and leaf margin was inherited together 'en bloc' under the control of a single dominant locus (the 'midrib complex') and was inseparably associated with the locus governing the purple coloration of the internode. The purple panicle was caused by a single dominant locus. Each of the three characters (purple lamina, purple stigma and purple seed) was governed by two complementary loci. One of the two loci...

Genome size and ploidy variations in pearl millet (Pennisetum glaucum) and napier grass (Pennisetum purpureum) genotypes Acta Agron. (Palmira)
Animasaun,David Adedayo; Morakinyo,Joseph Akintade; Mustapha,Oba Toyin; Krishnamurthy,Rama.
Abstract Pennisetum glaucum (Pearl millet) and P. purpureum (Napier grass) are economically important members of the genus Pennisetum . The knowledge of variability in chromosome number, size and genomic content of the species could provide clues on the mechanisms responsible for decrease or increase in genomic content in the evolutionary pathway. In the present study, twenty nine genotypes consisting of 24 pearl millet and 5 Napier grass were assessed for inter- and intra-variations in chromosome number, ploidy status and genome content. Conventional cytogenetic was used for chromosome counting and flow cytometry technique for assessing genomic contents of the genotypes. Pearl millet genotypes were diploid (2n = 14) while Napier grass were mainly...

Meiosis in elephant grass (Pennisetum purpureum), pearl millet (Pennisetum glaucum) (Poaceae, Poales) and their interspecific hybrids Genet. Mol. Biol.
Techio,Vânia Helena; Davide,Lisete Chamma; Pereira,Antônio Vander.
The cultivated and sexually compatible species Pennisetum purpureum (elephant grass, 2n = 4x = 28) and Pennisetum glaucum (pearl millet, 2n = 2x = 14) can undergo hybridization which favors the amplification of their genetic background and the introgression of favorable alleles into breeding programs. The main problem with interspecific hybrids of these species is infertility due to triploidy (2n = 3x = 21). This study describes meiosis in elephant grass x pearl millet hybrids and their progenitors. Panicles were prepared according to the conventional protocol for meiotic studies and Alexander’s stain was used for assessing pollen viability. Pearl millet accessions presented regular meiosis with seven bivalents and high pollen viability. For elephant...

Filardi,RS; Junqueira,OM; Casartelli,EM; Laurentiz,AC; Duarte,KF; Assuena,V.
An experiment was carried out to evaluate the effect of replacing corn with pearl millet in commercial layer diets, formulated according to the minimal requirements for total and digestible amino acids. Two hundred and forty Lohmann LSL laying hens with 25 weeks of age were distributed in a completely randomized experimental design according to a 2 x 5 factorial arrangement with 3 replicates of 8 birds. Feed was formulated on two amino acid basis (total or digestible) according to Rostagno et al. (2000) and there were five pearl millet inclusion levels (0%, 25%, 50%, 75%, and 100%). Performance and egg quality were evaluated during five periods of 21 days.At the end of each period, feed intake, egg production, egg weight and feed conversion were evaluated....

Production of polyclonal antisera to manganese superoxide dismutase expressed in downy mildew resistant pearl millet and its application for immunodiagnosis Electron. J. Biotechnol.
Urs Babitha,Mysore Pandurangaraj; Madhu,Devaiah; Sripathi P,Harishchandra; Shekar Shetty,Hunthrike.
Level of superoxide dismutase (SOD) in the seedlings of downy mildew resistant pearl millet genotype (IP 18293) increased 3-fold upon inoculation with Sclerospora graminicola. Mn-SOD was purified from pearl millet by ion-exchange and gel filtration chromatography. Polyclonal antibody was raised in rabbit against Mn-SOD of pearl millet. Anti-Mn SOD had an antibody titer of 1:20,000. ELISA results revealed a 1.5-fold increase in pure Mn-SOD preparation compared to the crude preparation. The antibody reactivity as detected by ELISA revealed that Mn-SOD is higher at 4 hrs post-inoculation in resistant pearl millet seedlings. The reactivity also showed the Mn-SOD is more in roots of inoculated resistant seedlings. SOD reactivity was also determined by DIBA...
